SARS-CoV2 detection on surface testing

Coronavirus SARS-CoV-2 (COVID-19 o 2019-nCoV) is proved to be persistent for even more than nine days on inanimate surfaces such as metal, glass and plastic, but it can be effectively inactivated through appropriate sanitisation (Kampf G. et al. Persistent of coronaviruses on inanimates surfaces and their inactivation with biocidal agents. Review. Journal of Hospital Infection. 2020). Scientific research is in continuous evolution.

The Tentamus Group based in Italy, through the Laemmegroup Laboratory, has introduced a new test to detect SARS-CoV2 on surfaces in relation to environmental monitoring.

The test is not for human or veterinary diagnostic use.

The method used is Real-Time PCR based on OMS suggested sequences to detect 2019-nCoV (causal agent of COVID-19), using extracts of RNA from environmental buffers. Due to the variability of tested samples, it is necessary to guarantee the inhibition of the PCR, the extraction yield in each experimental session and exclude false negative results, through a specific artificial process control resistant to nuclease.

The artificial process control related to the kit is the Intype IC RNA. The kit contains a 2-plex oligo mix that can be used on almost all Real-Time PCR instruments available on the market, combined with a high-quality one-step Rtqpcr mix.

In the reaction mixture there are two fluorophores: FAM for the detection of 2019-nCoV and HEX for the detection of Intype IC RNA. The kit allows to highlight the presence of virions in surfaces and fomites that in turn can facilitate the transmission of the virus from one person to another.

The result is provided in 48-72 hours from the moment the sample reaches the laboratory (timing will be agreed with your commercial agent and is intended as working days) and will be expressed n terms of present/absent, being a qualitative test.

The limit of determination (LOD) of the method is 3.8 genomic units.

The test is not accredited.

The sampling must be performed using the kit provided by the laboratory and in accordance with the instructions provided. No different materials are allowed.

Samples must be delivered to Laemmegroup Laboratory in appropriate temperature conditions and in the shortest time possible, not exceeding 24-48 hours from sampling.
